用途:

  GFP版的密閉式螢光成像系統在分子與細胞生物學的綠色螢光蛋白中可提供多種應用. 基本版 Closed FluorCam (CCD camera, 4 fixed LED panels) 再增加全機動與軟體控制的濾片輪 filter wheel 以及適當的濾片組來偵測與擷取GFP,  red-shifted GFP (EGFP), 或其他多種螢光蛋白畫面.

   此儀器在GFP 與葉綠素螢光偵測及擷取畫面間提供了友善快速的轉換. 此機非常精巧可讓樣品做暗適應. 價格包含4組升級的LED 光源板的FluorCam 控制單元、高解析度相機、已裝軟體的控制電腦以及濾片輪及濾片做GFP 與葉綠素螢光偵測.


主要特點:

  • Combined imaging of GFP and chlorophyll fluorescence.
  • Included high-resolution CCD camera: full resolution 1392 x 1040 (15 fps), 30 fps at resolution 640 x 480; integration times from microseconds to seconds; binning options. The camera is specifically intended for detection of weak signals demanding long integration times; the setup retains most of the functionalities for chlorophyll fluorescence kinetics measurement.
  • Four light-upgraded LED panels (4 panels 470 nm OR 2 panels 447 nm + 2 panels 470 nm OR 2 panels 627 nm and 2 panels 470 nm) .
  • Additional UV, green, yellow, amber, orange or infrared light source (optional).
  • 8 position filter wheel - motorized and software-controlled (included).

GFP常用濾片組:

Proteins of the GFP family demonstrate a great spectral diversity and they often have different excitation and emission spectra. Therefore it is important to use the most appropriate filter sets so as to obtain maximum sensitivity for FP viewing.

  • Red-shifted variant ( EGFP)
    The excitation peak is centered at about 488 nm and the emission peak is at 507 - 509 nm.
    Suggested filters: short-pass 480 nm for excitation and 532/28 or 530/25 for emission.
  • Original (wild type) GFP variant (wtGPF)
    The major excitation peak is centered at about 395 - 397 nm and the emission peak is at 504 nm.
    Suggested filters: short-pass 420 nm for excitation and 532/28 or 530/25 for emission.
  • Blue-emitting variant (BFP)
    The excitation peak at about 384 nm and the emission peak near 448 nm.
    Suggested filters: short-pass 400 nm for excitation and 469/35 for emission.
